752 Endothelial glycocalyx destruction is the main culprit for cutaneous inflammation in psoriasis

The skin vasculature ensures local homeostasis and plays an essential role in inflammatory diseases. Tortuous dilated vessels are one of the hallmarks psoriasis; however, how they participate psoriatic pathogenesis remains unclear. Here, we profiled single-cell transcriptomes vascular endothelial cells (ECs) from healthy discovered heterogeneity ECs signaling pathways immune responses. Pseudotime analysis demonstrated that capillary EC subsets were highly activated. Among most functional alterations psoriasis, noticed genes related to degradation glycocalyx, critical component barrier, upregulated. Transmission electron microscopy, immunofluorescence, ELISA further validated substantial destruction glycocalyx vessels. Of note, degrading IMQ-induced mice enhanced responses aggravated inflammation. Whole-mount immunofluorescence staining revealed vessel remodeling T cell infiltration by degradation.
